Decline of testosterone production with age has led to interest in androgen replacement therapy. As demonstrated by a meta-analysis, substitution therapy with testosterone results in a significant reduction of inflammatory markers. For women with PCOS, hormones like birth control pills can be used to help lessen the effects of this increased level of testosterone. In people who have undergone testosterone deprivation therapy, testosterone increases beyond the castrate level have been shown to increase the rate of spread of an existing prostate cancer. The brain is also affected by this sexual differentiation; the enzyme aromatase converts testosterone into estradiol that is responsible for masculinization of the brain in male mice.
The current analysis was part of a larger study assessing the effects of exogenous testosterone administration on changes in body composition after a 28-day exercise- and diet-induced energy deficit (ED) designed to be 45% of total energy needs (43). Few studies have considered the side effects of relaxin either in high or low levels. Women who have low progesterone levels and who do succeed in getting pregnant are at higher risk for miscarriage or pre-term delivery, because the hormone helps maintain the pregnancy.
Serum testosterone levels were similar between WT, 5 μg of AR-ASO, and Control-ASO- injected AR-97Q mice (Fig. 3e). A Anti-human androgen receptor (AR) immunostaining of motor neurons and skeletal muscles of male wild-type (WT), male AR-97Q, and female AR-97Q mice at postnatal day 1 (P1), P7, and testosterone (T)-administered female AR97Q mice at P4. Immunohistochemical analysis showed that AR also accumulated in the nucleus of motor neurons and skeletal muscle cells in male AR-24Q mice but not in female AR-24Q mice at P1 (Supplementary Fig. 1).
As for the hyperexcitability in SBMA, the elevation of REST target glutamate receptor genes may lead to excess glutamate uptake and excitotoxicity in motor neurons. These results suggest that motor neurons expressing mutant AR in early stages of disease in SBMA are hyperexcitable. Interestingly, REST negatively regulates the expression of Rest4 via silencing the expression of splicing factors, such as SRRM3 and SRRM452.
